Quantitative proteomics of cerebrospinal fluid using tandem mass tags in dogs with recurrent epileptic seizures

چکیده
This prospective study included four dog groups (group A: healthy dogs, B: dogs with idiopathic epilepsy under antiepileptic medication (AEM), C: without AEM administration, D: structural epilepsy). The purpose of the was to compare proteomic profile among groups. Samples were analyzed by a quantitative Tandem Mass Tags approach using Q-Exactive-Plus mass-spectrometer. Identification and relative quantification performed Proteome Discoverer, data R. Gene ontology terms based on Canis lupus familiaris database. Data are available via ProteomeXchange identifier PXD018893. Eighteen proteins statistically significant (P < 0.05). MMP2 EFEMP2 appeared down-regulated whereas HP APO-A1 up-regulated (groups B, D). CLEC3B PEBP4 C). IGLL1 C) only protein detected significantly different epileptic dogs. Western blot SPARCL immunoassay used quantify abundance change, validating analysis. Both, showed good correlation levels identified through analysis (r = 0.712 r 0.703, respectively). from CSF seizures could reflect that MMP2, may contribute blood-brain barrier disruption seizure-induced inflammatory process in brain. change indicate activation protective mechanisms within brain tissue. Antiepileptic influence several cellular responses alter proteome composition.

Efficient Interpretation of Tandem Mass Tags in Top-Down Proteomics
Mass spectrometry is the major analytical tool for the identification and quantification of proteins in biological samples. In so-called top-down proteomics, separation and mass spectrometric analysis is performed at the level of intact proteins, without preparatory digestion steps. It has been shown that the tandem mass tag (TMT) labeling technology, which is often used for quantification base...
متن کاملQuantitative proteomics of cerebrospinal fluid from patients with Alzheimer disease.
Biomarkers to assist in the diagnosis and medical management of Alzheimer disease (AD) are a pressing need. We have employed a proteomic approach, microcapillary liquid chromatography mass spectrometry of proteins labeled with isotope-coded affinity tags (ICAT), to quantify relative changes in the proteome of human cerebrospinal fluid (CSF) obtained from the lumbar cistern. Using CSF from well-...

Epilepsy is a common neurological disorder with seizures, but diagnostic approaches in veterinary clinics remain limited. Cerebrospinal fluid (CSF) is a body fluid used for diagnosis in veterinary medicine. In this study, we explored canine epilepsy diagnostic biomarkers using gas chromatography-mass spectrometry (GC-MS)-based metabolic profiling of CSF and multivariate data analysis. Profiles ...
متن کاملAlterations of endocannabinoids in cerebrospinal fluid of dogs with epileptic seizure disorder
BACKGROUND Epilepsy is one of the most common chronic neurological disorders in dogs characterized by recurrent seizures. The endocannabinoid (EC) system plays a central role in suppressing pathologic neuronal excitability and in controlling the spread of activity in an epileptic network. Endocannabinoids are released on demand and their dysregulation has been described in several pathological ...
